Abstract:Structural study on flexible, dynamic and heterogeneous macromolecules is challenging by current techniques, including X-ray crystallography, nuclear magnetic resonance (NMR) spectrum, cryo-electron microscopy (cryo-EM) single-particle reconstruction, due to requirement of averaging the signals from thousands to millions of different macromolecules that are assumed sharing a single structure. We reported an approach to determine the 3D structure of a single-instance macromolecule at near a nanometer resolution, termed individual-particle electron tomography (IPET). IPET does not require a pre-given initial model, averaging of multiple molecules or an extended ordered lattice, but can achieve a 3D structure at near nanometer resolution from heterogeneous sample via structure determination of each individual macromolecule. The comparison of these macromolecule structures provides an experimental opportunity to reveal macromolecular dynamics, equilibrium fluctuation, mechanism and even catching the intermediate-state 3D structure during chemical reaction. |
